Ubiquitin(Ub),ahighlyconserved,8kDapolypeptidepresentinalleukaryoticcells,isconjugatedtotheε-aminogroupoflysineresiduesinthetargetproteinthroughthesequentialactionofthreeenzymes,anE1Ubactivatingenzyme,anE2conjugatingenzyme,andanE3ligase.Inaddition,thesevenlysineswithinUbitselfcanserveasUbacceptorsleADIngtotheformationofpolyubiquitinchains.AmongtheseK48andK63linkagesarewellcharacterized.K48-linkedpolyubiquitylationtargetsproteinsforproteasomaldegradationwhereasK63linkagesregulatesignalingevents,receptorendocytosisandimmuneresponses.Polyubiquitinlinkagesatotherlysinesarelessprominentandtheirphysiologicalroleremainslargelyunknown.
VU-1isamonoclonalantibodythathasbeenshowntorecognizefreeubiquitin,K48-,K63-,K11-andlinearchainsonWesternblot.Otherlinkageshavenotbeenspecificallytested. VU-1isanexcellentantibodyforimmunostainingapplicationsgeneratingrobustubiquitindetectionandlowbackgroundwithanABIlitytodetectsubcellularubiquitinlocalization.
